膀胱移行细胞癌中环氧化酶-2的表达研究|透明细胞癌恶性程度

  摘要: 目的:检测环氧化酶-2(COX-2)在膀胱癌组织中的表达,探讨COX-2表达与膀胱癌临 床病理的关系。方法:采用免疫组化方法检测56例膀胱癌组织中COX-2的表达。结果:膀胱癌 组织中COX-2的阳性表达率为57.1%。COX-2的表达与肿瘤的病理分级和临床分期相关。结 论:COX-2的异常表达可能在膀胱移行细胞癌的发生发展中起一定的作用。

  Expression of cyclooxygenase-2 in human transitional cell carcinoma of t he bladder/LU Hao-yuan, BAI Xian-zhong∥The Affiliated Tumor Hospital of Guang xi Medical University, Nanning 530027, China
  Abstract:Objective:To study the expression of cyclooxygenase-2(COX-2) in human transiti onal cel l carcinoma of the bladder(BTCC)and their relationship with clinical pathologic al characteristics. Methods: S-P immunohistochemistry staining was performed t o e valuate the positivity of COX-2 in fifty-six cases of transitional cell carcin om a of the bladder and 10 cases of normal bladder mucosa. Results: The positive e xp ression of COX-2 were 57.1%(32/56), the expression of COX-2 was related tocl inic stage and the tumor grade(PTMplus广谱试剂盒(KIT-9901);用EDTA修复法修复 抗原;DAB法显色。采用已经研究证实为COX-2蛋白阳性表达的肺癌标本作为阳性对照,以P BS替代一抗为空白对照。
  1.3 结果判断
  参照文献报道[1],显微镜下观察细胞染色情况,COX-2阳性细胞的标志为细胞浆 内或胞膜 上呈棕黄色,分4个级别。分级标准为∶-,细胞无着色;+,细胞胞浆内微弱且弥漫染色 (可以包括90%的细胞胞浆内强染色;-,判定为阴性表达,+~�为阳性表达。
  1.4 统计学分析
  实验结果采用SPSS 10.0进行统计处理,α值取0.05作为显著性标准。
  
  2 结果
  
  2.1 COX-2在膀胱癌细胞的表达
  COX-2阳性表达产物呈黄色颗粒状,表达较强时呈棕黄色,定位于癌细胞浆中。COX- 2在膀胱癌细胞中阳性表达率为57.14%(32/56),其中弱阳性11例,中等阳性15例,强阳性 6例,但在正常膀胱细胞中为阴性表达,见表1。
  COX-2的阳性表达与膀胱移行细胞癌的生物学行为密切相关,与膀胱移行细胞癌病理分级、 临床分期密切相关,差异有统计学意义(分别为P
