【摘要】 目的 探讨孕鼠铅暴露对仔鼠血铅水平的影响。方法 wistar大鼠42只,雌雄各半,按雌雄配对分为五组(A、B、C、D、E)。A组为正常对照组,进食正常饲料B、C、D、E组每公斤饲料分别添加醋酸铅0.3 g、0.9 g、2.7 g、8.1 g,产仔后恢复正常饲料。母鼠与仔鼠组a~e 1、2于生后1、3周分别进行血铅测定。结果 仔鼠血铅水平与母鼠血铅水平呈正相关(r=0.517)。结论 胎鼠血铅水平与母鼠孕期铅暴露的程度相关。

A study of the impact ofzinc exposure of pregnant mice on zinc level in plasma of filial mice

【Abstract】 Objective To explore the impact of zinc exposure of pregnant mice on zinc level in plasma of filial mice. Methods 42 mice (a half male and a half female), grouped into five groups, were used in the experiment. Group A, the control group was offered normal food, while Groups B, C, D and E were offered food containing acetic zinc 0.3 g, 0.9 g, 2.7 g and 8.1 g per kilo food.The zinc levels in plasma of the parent and filial mice in groups A-E were tested three weeks after the birth of the filial mice. Results The zinc levels in plasma of the filial mice were positively correlated with that of the parent mice (with a correlation coefficient of r=0.517). ConclusionThe zinc level in plasma of filial mice is correlated with the extent to which the parent mice expose to zinc pollution during pregnancy.

铅是一种有毒的重金属,又广泛存在于大自然中。铅可通过胎盘屏障进入胎儿体内。为探讨孕期铅毒对雌鼠生长期及孕期体质量的影响及血铅对后代血铅水平的影响程度,笔者对wistar大鼠添加不同剂量的醋酸铅喂养,母鼠产仔后与仔鼠进行血铅水平检测,报告如下。
1 材料及方法
1.1 材料 动物模型:wistar大鼠42只,雌雄各半,雄性体质量200~250 g,雌性体质量160~180 g(成都军区驻昆总医院动物培养中心提供),正常饲料喂养一周后,按雌雄配对分为五组(A、B、C、D、E)。根据文献[1],饲料配方为:A组进食正常饲料,B、C、D、E组每公斤饲料分别添加醋酸铅0.3、0.9、2.7、8.1 g,待产仔后恢复正常饲料。实验期观察其体质量、毛发、活动等变化,并做记录。仔鼠出生后,按A、B、C、D、E各组所产仔鼠分为两组,设定为a1/a2,b1/b2,c1/c2,d1/d2,e1/e2,分别于出生第1周和第3周进行血铅测定。
1.2 方法
1.2.1 血铅测定 一周龄仔鼠断头取血,三周龄仔鼠及成鼠心脏穿刺取血,加入肝素抗凝试管内备查,血铅检测应用美国PerkinElmer公司产AAnalyst-300Atomic Absorption Spectromete(PE-300)石墨炉原子吸收光谱仪,血铅测定按照标准方法进行[3],按卫生部质量保证要求进行质量控制。
1.2.2 统计学处理 统计分析应用医学百科全书、医学统计软件(PEMS)进行计量资料描述,两样本比较t检验及秩合检验。
2 结果
2.1 雌鼠孕期体质量检测结果 饲养过程中,观察各组活动度、毛发等状况未见异常,雌雄鼠体质量随周龄的增加而增加,各雄鼠不同组间,体质量比较无显著性差异(P
